This preprint investigates the factors that determine whether a macrophage will phagocytose or trogocytose a target cell. The authors found that target cell cortical tension is a key regulator of this process, with macrophages choosing trogocytosigs of cells with low tension and phagocytosis of cells with high tension.

This preprint demonstrates that the affinity maturation of antibodies is driven by differential plasma cell proliferation, rather than selective differentiation, after they exit the germinal center.

This finding challenges current models of plasma cell differentiation and provides key insights into how high-affinity antibody-producing cells are selected for in the immune response.

A first-of-its-kind characterization of T cell subsets in the murine mammary gland, which undergoes massive remodeling during periods of pregnancy and lactation. Most notably, the authors spotted a subset of intraepithelial-like T cells (IELs), which expanded during lactation and had originated in the gut (as opposed to the thymus!). Mammary IELs were conserved in human breast tissue and milk and are thought to act as a first line of defense when the mammary gland acts as a temporary barrier tissue during breastfeeding.

This preprint is relevant because it explores how herpes simplex virus type 1 (HSV-1) infection remodels the mechanical properties of the cell nucleus, finding that the nucleus softens during infection. This softening is linked to changes in nuclear components such as the viral replication compartment, nuclear lamina, and forces acting on the nucleus.

This preprint highlights the functional specialization of dermal macrophages in the ear skin. While transcriptional heterogeneity of resident macrophages across tissues has been appreciated, this paper adds to that on the intra-tissue scale. It demonstrates that cues from the tissue microenvironment as opposed to cellular origin shape macrophage profiles – allowing to fine-tune responses from a generally pro-homeostatic tissue-resident and a pro-inflammatory monocyte-derived macrophage population. The authors illustrate this with multidimensional transcriptomic data in the physiological context of mycobacterial infection using intradermal BCG injection and Irf8-/-driven monocytopenia.

This preprint shows miR-130b as a key regulator of B cell tolerance, which is critical for preventing autoimmune diseases, and demonstrates that ERα is essential for maintaining adequate control of central B cell tolerance. A type 2 immune bias has been associated with pregnant and neonatal individuals. This preprint explores how maternal type 2 immunity, via IL-4 receptor signaling, directly translates to their offspring. The authors attribute maternal microchimerism – the transfer of maternal (immune) cells into offspring – to play a key role in this process. Murine pups fostered by IL-4 receptor-deficient mothers exhibited fewer maternal cells and a reduced type 2 bias, which would increase their susceptibility to early worm infections.

This paper examines the ability of various B cell subsets to engulf and process antigens, revealing that while Follicular B, marginal zone, and B1 cells all engulf targets via their B cell receptors, only B1 cells in the peritoneal cavity can engulf non-cognate antigens. However, the study also shows that B1 cells don’t effectively activate T cells or kill bacteria, whereas Follicular B cells efficiently present engulfed antigens to T cells.

Peer Review in Biomedical Sciences

Communication of scientific knowledge has changed dramatically in recent decades and the public perception of scientific discoveries depends on the peer review process of articles published in scientific journals. Preprints are key vehicles for the dissemination of scientific discoveries, but they are still not properly recognized by the scientific community since peer review is very limited. On the other hand, peer review is very heterogeneous and a fundamental aspect to improve it is to train young scientists on how to think critically and how to evaluate scientific knowledge in a professional way. Thus, this course aims to: i) train students on how to perform peer review of scientific manuscripts in a professional manner; ii) develop students' critical thinking; iii) contribute to the appreciation of preprints as important vehicles for the dissemination of scientific knowledge without restrictions; iv) contribute to the development of students' curricula, as their opinions will be published and indexed on the preLights platform. The evaluations will be based on qualitative analyses of the oral presentations of preprints in the field of biomedical sciences deposited in the bioRxiv server, of the critical reports written by the students, as well as of the participation of the students during the preprints discussions.
