Cell signalling and mechanosensing
Guidance of cellular nematics into shape-programmable living surfaces

In this preprint, the authors have managed to organise a flat sheet of cells in such a way that, when this cell sheet detaches from an adherent surface, it automatically/naturally folds into a preprogrammed 3D shape. This is possible due to a specific arrangement of cells, where the neighboring cells are locally aligned parallel to each other, while on a global scale, this alignment creates patterns of tissue-scale tensile forces, which then dictate how the detached cell sheet will fold to form various structures in 3D. The authors have achieved a variety of 3D shapes using different arrangements of cells, which are in turn achieved through different patterns of surface coating where cells attach.

Genetically encoded mechano-sensors with versatile readouts and compact size

Long-range chemical signalling in vivo is regulated by mechanical signals

Axonal pathfinding is the process that allows various neuronal cells to make connections with their target cells. Axons use a variety of cues to find their path towards their target, though how this process is regulated is still an open question. In this context, the authors of this preprint investigate the pathfinding in retinal ganglion cells. They find that the mechanosensitive ion channel Piezo1 is required for proper expression and interpretation of guidance cues such as Semaphorin3A and Slit1. The preprint also finds a role for tissue stiffness (regulated by Piezo1) in guiding the migrating axons.

JAK2 signalling to HNRNPA1 represses retrotransposon activity in haematopoietic stem cells

Random genomic insertion of active transposons facilitates genetic variation. However, unhindered transposon activity can have deleterious consequences. This preprint identifies a new signalling pathway downstream JAK2 signalling to suppress transposon activity in haematopoeitic stem cells.

LIM Domain Proteins link molecular and global tension by recognizing strained actin in adhesions

It feels intuitive that global tension must translate to local/molecular tension in a cell, especially in known mechanosensitive proteins, that are recruited at cell-ECM and cell-cell adhesion sites. The authors of this preprint demonstrate that this is not the case, and that specific mechanosensitive proteins can distinguish the two. LIM domain proteins such as zyxin recognise the global tension, while the known mechanosensitive protein Vinculin seems to only respond to molecular tension and not to global tension in a cell.

A mitotic bookmark coordinates transcription and replication

How transcription and DNA replication are coordinated in fast-replicating nuclei is poorly understood. In this preprint, the authors focus on the rapid nuclear division cycles during the syncytial blastoderm stage, where the zygotic genome is progressively activated for transcription, in parallel with DNA replication. The authors find a role for Brd4 as a bookmark to identify genes that are being transcribed, so that the Cdc7 activity will replicate them first, freeing up these loci for transcription, to minimise the conflict between DNA replication and transcription. DNA replication dislodges these bookmarks, which are then reestablished during the next cycle due to transcription.

Adaptive cytoskeletal responses to extracellular environment viscosity modulate cell migration

How the mechanical properties of the extracellular environment affect the ability of the cells to migrate is an open question. The authors focus on this question in the context of cancer metastasis and probe for the effects of extracellular viscosity. In this preprint, the authors show that extracellular viscosity modulates cancer cell migration by regulating actin dynamics and adaptation of adhesion.

Mechanical stress and caveolin-1 control the release of extracellular vesicles with increased tumorigenic properties

Release of extracellular vesicles (EVs) that carry cargo that remodels the extracellular matrix is one method cancer cells use to enable metastasis. This study reveals that release of EVs is modulated by mechanical stress. Caveolae, invaginations in the membrane that act as mechanosensors, are responsible for this effect.

Capillary constrictions prime cancer cell tumorigenicity through PIEZO1

By mimicking the physiological architecture of microcapillaries with a microfluidic device, the authors investigate how physical constraints affect melanoma cells under a specific fluid flow rate, potentially surpassing the limitations of in vivo models. They show that changes in capillary diameters not only trigger chromatin remodeling but also promote cancer stem cell-like phenotypes, characterized by altered gene expression levels and invasiveness conveyed through Piezo1-mediated mechanosensing.

Recognition of small Tim chaperones by the mitochondrial Yme1 protease

This preprint contibutes to understanding the so far rather elusive mechanisms of how mitochondrial AAA+ proteases recognise their substrates. They show that the Yme1L protease in yeast surveils the folding state of its substrate Tim10 by always binding to it, independent of its conformation, but only degrades Tim10 when the protein is unfolded. Interestingly, the assembled Tim10-Tim9 chaperone complex, which is not a degradation target, is still “screened” by Yme1L but uses a different protein interface to bind Yme1L compared to the Tim10 monomer.

Scalable automated segmentation quantifies mitochondrial proteins and morphology at the nanoscale

Peer Review in Biomedical Sciences
Communication of scientific knowledge has changed dramatically in recent decades and the public perception of scientific discoveries depends on the peer review process of articles published in scientific journals. Preprints are key vehicles for the dissemination of scientific discoveries, but they are still not properly recognized by the scientific community since peer review is very limited. On the other hand, peer review is very heterogeneous and a fundamental aspect to improve it is to train young scientists on how to think critically and how to evaluate scientific knowledge in a professional way. Thus, this course aims to: i) train students on how to perform peer review of scientific manuscripts in a professional manner; ii) develop students' critical thinking; iii) contribute to the appreciation of preprints as important vehicles for the dissemination of scientific knowledge without restrictions; iv) contribute to the development of students' curricula, as their opinions will be published and indexed on the preLights platform. The evaluations will be based on qualitative analyses of the oral presentations of preprints in the field of biomedical sciences deposited in the bioRxiv server, of the critical reports written by the students, as well as of the participation of the students during the preprints discussions.
