This work focuses on a regulator of mitotic spindle organization called Nuclear Mitotic Apparatus (NuMA), which the authors show to be responsive to external mechanical stimuli. This preprint adds important details to our understanding of what regulates the in-plane divisions in epithelial tissues, even though the apicobasal axis is the long axis of the cell. The authors distinguish the role of cell elongation vs. mechanical forces in the plane of the epithelium to orient the spindle.

Chromosomal missegregation and aneuploidy during cell division are hallmarks of cancerous cells. In this preprint, the authors show that Wnt signalling, specifically that initiated by Wnt10b, is involved in regulating cell division during the S-phase. During DNA replication stress, Wnt10b signalling prevents premature microtubule dynamics, reducing replication-induced chromosomal breaks.

This preprint focuses on microtubules made of distinct alpha-tubulin isotypes, and how they regulate binding of distinct motor proteins and Microtubule Associated Proteins (MAPs). Their experiments in budding yeast provide insights into spindle morphogenesis, as the combined activity of the two isotypes (Tub1 and 3) is required to build a spindle with normal length.

During oogenesis, early oocytes must become polarised. This study identifies kinesin-1 and ensconsin (MAP7) as components that facilitate recruitment of microtubule organising centres (MTOC) to the cortex to allow polarisation. Kinesin-1/ensconsin-mediated recruitment of MTOC factors may be a mechanism utilised in multiple cell types to establish polarity.

Endoplasmic Reticulum (ER) exists as sheets and tubules, responding to reorganization of the cytoskeleton. Such reorganization can happen during cell migration and wound healing. The authors show that ER reorganization is an essential component of a cell’s response to physical stimuli, as it regulates the orientation of the mature focal adhesions.

Nuclear envelope budding is a non-canonical mechanism to export large transcripts in muscle cells

In this preprint, the authors describe a non-canonical pathway for nuclear export of extremely large transcripts, which can be best described as transport between the inner and outer membranes of the nuclear envelope using an inverse of a neuronal synapse. This is especially required in muscles, as muscles contain several large proteins, including one of the largest proteins, Titin. This pathway is specifically activated as muscle cells differentiate from myoblasts to myotubes, and resembles the pathway used by the Herpesvirus to similarly export itself from the nucleus.

Inverted Assembly of the Lens Within Ocular Organoids Reveals Alternate Paths to Ocular Morphogenesis

This exciting preprint highlights the emergence of lens-like tissues from ocular organoids derived from medaka pluripotent embryonic stem cells, developing in an “outside-in” fashion. Consistent with the in vivo positioning of the lens surrounded by cup-shaped retinal tissue, this finding not only suggests the possibility of an alternative mechanism for lens formation but also demonstrates the potential of organoids as a promising tool to study cell and tissue organization and observe diverse modes of morphogenesis.

Compartmentalization of mRNA and Translation by Desmosomes

This preprint focuses on the spatial (re)organization of translation machinery. Desmosomes, which are part of epithelial septate junctions, are shown here to be involved in regulating the translation of cortical mRNAs. This adds another method for the local regulation of translation. The authors show that the transcripts associated with GO terms, cell adhesion and cytoskeleton, were enriched at desmosomes, but only during wound healing, suggesting that these transcripts are maintained as an inactive reservoir to mount a quick response to wounding and to quickly recover epithelial barrier function.

Fibroblast growth factor 21 regulates neuromuscular junction innervation through HDAC4 in denervation-induced skeletal muscle atrophy

The preprint teases apart the role of FGF21, a muscle-secreted paracrine/autocrine signal, that is responsible for muscular atrophy. The secretion is normally triggered in response to a loss of innervation, but an artificial upregulation of FGF21 can also feedback and denervate the skeletal muscles. Downregulating FGF21 can also block denervation and prevent muscular atrophy, suggesting a promising strategy to mitigate skeletal muscle atrophy.

Cells remodel their actin cytoskeleton in response to mechanical stimuli. This preprint identifiy Solo (RhoGEF) regulation on PDZ-RhoGEF (PRG) and leukemia-associated RhoGEF (LARG) during mechanical stress and is also required to maintain LARG activity, which in turn is necessary for Solo-induced actin polymerization. This indicates PRG/LARG function cooperatively in Solo-mediated actin regulation in response to substrate stiffness.

Vibha: This preprint describes a single-component optogenetic system exploiting an light controlled talin homodimerization, overcoming limitations of previous dual-component systems. Optogenetic dimerization of talin boosts adhesion formation and signaling, whereas monomerization interrupts these processes, emphasizing the crucial role of talin dimerization in cell-ECM interactions.

Girish: This preprint describes new tools to tune cell-ECM adhesion via controlling the dimerization of Talin, a component of integrin-based cell-ECM adhesion. Talin naturally exists as a dimer in its active state. Replacing its dimerization domain with a light-inducible dimerization-dissociation fluorescent protein allowed the authors to regulate dimerization directly, controlling the activity of Talin and allowing a thorough characterization of this new tool.

This preprint describes optogenetic tools to activate core cellular pathways in a developmental context. The tools described can activate FGF, Nodal, and BMP signaling pathways, with spatiotemporal specificity. The authors exploit the dimerization of the LOV domain to activate these pathways, as a common theme between these pathways is the dimerization of their signalling components. The authors claim that such an optogenetic strategy can be a more general one, and could likely be implemented in various signaling contexts that involve such dimerization.

This cool study analyzes a large collection wild nematodes to find evidence of mitonuclear co-evolution. The authors subsequently used a neat genetic trick to generate different combinations of mitochondrial and nuclear genome and found strong mitonuclear epistasis in many cases.

This preList contains preprints discussed during the Metabolic and Nutritional Control of Development and Cell Fate Keystone Symposia. This conference was organized by Lydia Finley and Ralph J. DeBerardinis and held in the Wylie Center and Tupper Manor at Endicott College, Beverly, MA, United States from May 7th to 9th 2024. This meeting marked the first in-person gathering of leading researchers exploring how metabolism influences development, including processes like cell fate, tissue patterning, and organ function, through nutrient availability and metabolic regulation. By integrating modern metabolic tools with genetic and epidemiological insights across model organisms, this event highlighted key mechanisms and identified open questions to advance the emerging field of developmental metabolism.

Peer Review in Biomedical Sciences

Communication of scientific knowledge has changed dramatically in recent decades and the public perception of scientific discoveries depends on the peer review process of articles published in scientific journals. Preprints are key vehicles for the dissemination of scientific discoveries, but they are still not properly recognized by the scientific community since peer review is very limited. On the other hand, peer review is very heterogeneous and a fundamental aspect to improve it is to train young scientists on how to think critically and how to evaluate scientific knowledge in a professional way. Thus, this course aims to: i) train students on how to perform peer review of scientific manuscripts in a professional manner; ii) develop students' critical thinking; iii) contribute to the appreciation of preprints as important vehicles for the dissemination of scientific knowledge without restrictions; iv) contribute to the development of students' curricula, as their opinions will be published and indexed on the preLights platform. The evaluations will be based on qualitative analyses of the oral presentations of preprints in the field of biomedical sciences deposited in the bioRxiv server, of the critical reports written by the students, as well as of the participation of the students during the preprints discussions.
