Hippo-activated cells induce non-cell autonomous tumorigenesis in Drosophila

The Hippo-pathway has been generally thought of as a tumour suppressor pathway, with inhibition of Hippo signalling promoting tumorigenesis. However, a few studies indicate that this function might be context-dependent. In this preprint, the authors find a similar context dependency, such that the activation of the Hippo pathway makes cells behave like “oncogenic niche cells”, non-autonomously inducing tumour-like markers in neighbouring cells.

Epigenetic remodeling via HDAC6 inhibition amplifies anti-tumoral immune responses in pediatric AML

This study finds that impairment of histone deacetylase 6 (HDAC6) slows the progression of myeloid leukaemia. The authors determine that upon its inhibition, chromatin around an immune modulatory gene encoding RNase T2 becomes more accessible, and several other immune modulators are upregulated. As such, HDAC6 is identified as a potential therapeutic target for myeloid leukaemia.

Actin crosslinking is required for force sensing at tricellular junctions

Tricellular junctions in focus! As the name suggests, tricellular junctions are points in the epithelial tissues where 3 cells make contact. The special geometry of these contacts makes their dynamics that much more interesting. Both of the preprints below focus on the dynamics of tricellular junctions in various epithelial systems in Drosophila, where morphogenetic movements are studied extensively.
The first preprint focuses on F-actin dynamics at the tricellular junctions in the embryonic ectoderm, to show that F-actin crosslinking (mediated by Fimbrin) is necessary for the stability of tricellular junctions. The authors show that the crosslinked F-actin is better suited to sustain mechanical forces and is required for adhesion stabilization through recruitment of various accessory proteins to cell-cell adhesion complexes, strengthening adhesion under tension.

Condensation of Sidekick at tricellular junctions organizes mechanical forces for cell-cell adhesion remodeling

The second preprint focuses on a tricellular adhesion protein called ‘Sidekick’ and shows that it undergoes phase separation to form condensates. These condensates are also known to regulate the dynamics of myosin-II, regulating this force-generating machinery during morphogenetic movements. The authors show that certain conserved motifs in the intracellular domain of Sidekick are responsible for its phase separation, and that mutating these motifs leads to disruption of myosin-II dynamics to cause defects in cell rearrangements, as a consequence of the disrupted condensation.

Filopodia numbers impact chemotactic migration speed

This preprint highlights the critical role of an optimal number of filopodia in the speed of ameboid cell migration, revealing a nonlinear relationship with migration speed. Notably, the absence of filopodia does not impair directed migration; rather, it slows it down.

Vacuolar protein sorting-associated protein 1 (Vps1) has membrane constricting and severing abilities required for endosomal protein trafficking

Development of the axonal βII-spectrin periodic skeleton requires active cytoskeletal remodelling

The membrane-associated periodic skeleton (MPS) consists of a regularly-spaced actin rings linked by spectrins that span neurites. The authors of this preprint reveal that significant cytoskeletal remodelling is required for its establishment.

PINK1 SUMOylation regulates basal mitophagy

Sex-dependent neural plasticity in response to damage

This work shows the fly gut as a simplified model to study damage-induced neural plasticity, illustrating a reversible increase in neural tissue. This plasticity exhibits sexual dimorphism, occurring exclusively in females and suppressed by neuronal masculinization, positioning it as a system to investigate sex-specific mechanisms of neural repair.

Embryonic Stem Cell-Specific Responses to DNA Replication Stress

Compared to other cell types, stem cells display a low rate of mutation. Stem cell-specific mechanisms to achieve this however are not well understood. This study proposes that mouse embryonic stem cells circumvent propagation of mutations by undergoing cell death should DNA crosslinks not be repaired in S phase by homology-directed mechanisms.

Structural evidence for metal ion catalysis in the ribosome

This study provides evidence for a catalytic metal ion residing in the peptidyl transferase centre of the ribosome, most likely Mg2+. The ion can be detected in a newly determined structure as well as when re-examining already available structures of ribosomes from different domains of life.

Epithelial fusion is mediated by a partial epithelial-mesenchymal transition

How epithelial tissues fuse during development is an open question. In this preprint, the authors show that the cells on the edges of the tissue boundary go through a partial mesenchymal-like state, allowing the tissues to fuse. This mesenchymal-like state can be characterised by a shift in cell polarity, morphology, and transcriptional profile, transitioning away from epithelial to mesenchymal-like characters.

Peer Review in Biomedical Sciences
Communication of scientific knowledge has changed dramatically in recent decades and the public perception of scientific discoveries depends on the peer review process of articles published in scientific journals. Preprints are key vehicles for the dissemination of scientific discoveries, but they are still not properly recognized by the scientific community since peer review is very limited. On the other hand, peer review is very heterogeneous and a fundamental aspect to improve it is to train young scientists on how to think critically and how to evaluate scientific knowledge in a professional way. Thus, this course aims to: i) train students on how to perform peer review of scientific manuscripts in a professional manner; ii) develop students' critical thinking; iii) contribute to the appreciation of preprints as important vehicles for the dissemination of scientific knowledge without restrictions; iv) contribute to the development of students' curricula, as their opinions will be published and indexed on the preLights platform. The evaluations will be based on qualitative analyses of the oral presentations of preprints in the field of biomedical sciences deposited in the bioRxiv server, of the critical reports written by the students, as well as of the participation of the students during the preprints discussions.
