Considering how XMAP215 cooperates with the gammaTubulin Ring complex to nucleate and establish microtubule arrays is poorly understood, this preprint is a significant step in understanding how such arrays are established.

Functional rescue of a fatal ERAD mutation via alternative splicing

This study shows that a pathogenic mutation in the ER-associated degradation (ERAD)-protein SEL1 can be rescued through alternative splicing which bypasses the mutation and results in a shorter, but functional, SEL1 protein. It also proposes exon-skipping as a therapeutic strategy and demonstates its effectiveness in patient-derived fibroblasts.

The mechanistic basis of cargo selection during Golgi maturation

The authors supply mechanistic evidence for the function of COPI in both Golgi-protein retention within trans-cisternae and recycling of escaped ER proteins from cis-cisternae. By solving the structure of the COPI coatamer with the cargo-adapter GOLPH3, they identify binding regions for Golgi-resident proteins adjacent to the previously proposed PI4P binding sites of GOLPH3. The structure also suggests that GOLPH3 binding to COPI blocks one of the binding sites for the K(X)Kxx signal present on ER-escaped proteins.

Dynamic regulation of the COPII interactome and collagen trafficking by site-specific glycosylation of Sec24D

The authors of this preprint characterised two O-GlcNAcylation sites in the intrinsically disordered region of Sec24D. They find that both sites are required for collagen secretion, but while one is glycosylated constitutively, the other is only temporarily glycosylated during collagen transport. The constitutive GlcNAcylation facilitates binding to myoferlin, a factor that may promote fusion between ER- and Golgi membranes, allowing the large collagen cargo to be trafficked.

β-Catenin is known to play a dual function in cells: being involved in both cell-cell adhesion and Wnt signaling. The Drosophila Synaptotagmin-like protein, Bitesize, is known for its function in organizing F-actin during early morphogenesis. In this preprint, the authors describe the Bitesize function at even earlier stages, during the syncytial development. At this stage, Bitesize is required to maintain the integrity of the pseudocleavage furrow, facilitating the nuclear divisions.

Peer Review in Biomedical Sciences

Communication of scientific knowledge has changed dramatically in recent decades and the public perception of scientific discoveries depends on the peer review process of articles published in scientific journals. Preprints are key vehicles for the dissemination of scientific discoveries, but they are still not properly recognized by the scientific community since peer review is very limited. On the other hand, peer review is very heterogeneous and a fundamental aspect to improve it is to train young scientists on how to think critically and how to evaluate scientific knowledge in a professional way. Thus, this course aims to: i) train students on how to perform peer review of scientific manuscripts in a professional manner; ii) develop students' critical thinking; iii) contribute to the appreciation of preprints as important vehicles for the dissemination of scientific knowledge without restrictions; iv) contribute to the development of students' curricula, as their opinions will be published and indexed on the preLights platform. The evaluations will be based on qualitative analyses of the oral presentations of preprints in the field of biomedical sciences deposited in the bioRxiv server, of the critical reports written by the students, as well as of the participation of the students during the preprints discussions.
