VDAC isoforms play distinct roles in modulating metabolism and maintaining mitochondrial function

Individual knock-outs of all three isoforms of the mitochondrial outer membrane channel VDAC in HeLa cells show that each isoform has a different impact on the mitochondrial proteome and on cellular metabolism. Surprisingly, only VDAC3 promotes mitochondrial respiration.

Desmoplakin tail domain position in the desmosomal plaque is isoform dependent

This preprint looks at the influence of different desmoplakin isoforms on desmosome organisation. These isoforms have different lengths, due to differences in lengths of their Rod domains. Rod domain length has been suspected to influence protein function, e.g. of the spectraplakin family, and this preprint provides some answers in this regard.

This preprint emphasizes the crucial role of fibulin-1, a basement membrane (BM) component, which, through its interaction with collagen type IV, supports the spermatheca, a structure that undergoes repeated cycles of BM stretching and expansion during ovulation. Dynamic regulation of Sec24C by phosphorylation and O-GlcNAcylation during cell cycle progression

The authors identify changes in post translational modifications of the COPII coat protein Sec24C upon mitotic entry. These modifications relocalise Sec24C away from ER exit sites and therefore likely play a role in halting secretion and aiding equal distribution of the endomembrane system between daughter cells during division.

Polarity reversal of stable microtubules during neuronal development

Keratins coordinate tissue spreading by balancing spreading forces with tissue material properties

The authors identify a keratin-mediated mechanism regulating tissue spreading in zebrafish embryos. During epiboly, actomyosin-driven stress build-up triggers keratin network accumulation, increasing viscosity to maintain tissue integrity and balance forces within the spreading tissue.

Pex30-dependent membrane contacts sites maintain ER lipid homeostasis

This preprint characterises previously enigmatic domains of the yeast ER-resident protein Pex30 found at ER membrane contact sites with various other organelles. Using a combination of in vivo, in vitro and in silico experiments, the authors show that Pex30 is required for phosphatidic acid homeostasis and identify phosphorylation sites that mediate membrane contact site formation based on the metabolic state of the cells.

The neuron-intrinsic membrane skeleton is required for motor neuron integrity throughout lifespan

The authors of this preprint study the requirement of the membrane periodic skeleton in C. elegans motor neurons throughout their life. They find that loss of its integral component, Beta-Spectrin, at all stages of life causes axon breakage and degeneration, revealing a new means to screen for factors required for axon degeneration and maintenance.

Genetically encoded reporters of actin filament organization in living cells and tissues

Existing actin probes only poorly represent the organisation of actin filaments. In this preprint, the authors generate new probes that allow distinction of actin filaments via fluorescence polarisation microscopy. This marks a significant advance in actin visualisation.

ERM proteins support perinuclear actin rim formation

This preprint investigates how ERM proteins link actin filaments to the nuclear envelope, which is important for cell migration, nuclear positioning, and transcriptional control. This study reveals a direct relationship between fluorescence excitation and cell mechanical properties, highlighting that the stiffness of T cells can be rapidly increased by exposing to excitation light. Also propose “photostiffening” as a new method for rapidly quantifying phototoxicity.

Peer Review in Biomedical Sciences
Communication of scientific knowledge has changed dramatically in recent decades and the public perception of scientific discoveries depends on the peer review process of articles published in scientific journals. Preprints are key vehicles for the dissemination of scientific discoveries, but they are still not properly recognized by the scientific community since peer review is very limited. On the other hand, peer review is very heterogeneous and a fundamental aspect to improve it is to train young scientists on how to think critically and how to evaluate scientific knowledge in a professional way. Thus, this course aims to: i) train students on how to perform peer review of scientific manuscripts in a professional manner; ii) develop students' critical thinking; iii) contribute to the appreciation of preprints as important vehicles for the dissemination of scientific knowledge without restrictions; iv) contribute to the development of students' curricula, as their opinions will be published and indexed on the preLights platform. The evaluations will be based on qualitative analyses of the oral presentations of preprints in the field of biomedical sciences deposited in the bioRxiv server, of the critical reports written by the students, as well as of the participation of the students during the preprints discussions.
