These research advances our understanding of the oncogenic functions of Lbc GEFs and provide a mechanistic framework for how cell contraction dynamics contribute to tumor invasion.

Receptor tyrosine kinase AXL regulates Golgi organization and function through an adhesion-Arf1 signaling axis in breast and lung cancers

Cell-matrix adhesion influences Golgi organization, and its disruption in cancer cells affects membrane trafficking and cargo processing. A screen identified different Golgi organization patterns in breast and lung cancer cells (MDAMB231 vs MCF7, A549 vs CaLu1), regulated by adhesion. In silico analysis linked receptor tyrosine kinase, AXL, to Golgi regulation. These findings validate AXL-Arf1 crosstalk as a key mediator of Golgi organization and function in cancer cells.

Sristi: This study is the first to capture the migratory behavior of mammalian primordial germ cells (PGC) from their origin to their target destination, the future gonad. As they traverse diverse cellular environments, PGCs encounter a variety of ECM conditions and physical constraints. Adapting to these changing surroundings, they exhibit dynamic changes in nuclear shapes, while maintaining a consistent migration strategy.

Tubulin isotypes polymerise into sectioned microtubules that locally regulate protein binding

There are multiple genes encoding different tubulins, but how these different isoforms affect microtubule structure and behaviour isn’t fully understood. This study reveals that co-polymerisation of two different tubulin isoforms causes sectioning of microtubules with differing protofilament numbers and causes differential binding of microtubule-associated proteins.

Quantitative intra-Golgi transport and organization data suggest the stable compartment nature of the Golgi

The mechanism of intra-Golgi secretory transport remains unclear, with the cisternal progression and stable compartment models under debate. In this study, the researchers utilize advanced Golgi imaging in nocodazole-induced ministacks, and report cargo transport velocity decreases from the cis to trans side and varies among different cargos. These findings challenge the cisternal progression model and support the stable compartment hypothesis for Golgi organization.

Mitochondrial function regulates cell growth kinetics to actively maintain mitochondrial homeostasis

This work focuses on the scaling of mitochondria with cell size and its implications in growth and division. Using a microfluidic yeast ‘mother machine’, they tracked the growth of yeast cells and mitochondrial volume over several generations. They shed light on how mitochondrial volume dictates the time of cell division, linking cellular growth rates to the homeostasis of mitochondrial volume.

Lysosome positioning affects NLRP3 inflammasome activation

This preprint shows a novel link between lysosome positioning and NLRP3 inflammasome activation, indicating that positioning lysosomes at the cell periphery increases inflammasome activation in response to nigericin in macrophages. The study highlights the influence of prolonged mechanical shear on fibroblasts inducing FMT (Fibroblast-to-myofibroblast transition). Regulation of plasma membrane tension through hydrostatic pressure and actin protrusion forces

The plasma membrane serves as a crucial signalling hub, with its mechanical tension influencing both membrane trafficking and signalling activity. However, the mechanisms regulating membrane tension remain unclear. Findings from this study reveal that the actin cytoskeleton regulates membrane tension through two mechanisms, (1) modulating intracellular hydrostatic pressure. (2) adjusting polymerization forces in actin-rich protrusions. Furthermore, authors highlight discrepancies between bare and apparent membrane tension, demonstrating that tether forces do not reliably reflect in-plane membrane tension.

Expression of transposable elements (TEs) has numerous detrimental effects, including DNA damage and an aggressive immune response, so is actively repressed. In embryonic stem cells (ESCs), however, TE expression is loosely allowed. How then do ESCs escape the negative consequences of TE expression? In this study, the authors find multiple mechanisms that suppress the immune response following TE expression, allowing normal embryonic development.

Three-dimensional cellularization in chytrid fungi uses distinct mechanisms from those driving one- and two-dimensional cytokinesis in animals and yeast

This preprint focuses on cellularization in Chytrid fungi. These fungi don’t proliferate through simple division. Instead, they go through nuclear division and dispersal in 3D volume, followed by cellularization that separates and creates a cell aggregate, which then disperses. The process of cellularization has similarities to cell division (1D cellularization), and cellularization in, say, Drosophila (2D cellularization), while key differences exist, which this manuscript explores.
