Can slowing ageing begin in the gut?
We are, collectively, obsessed with not getting old. The global longevity market involves billions of dollars, serum ingredients cycle through trend cycles, and a growing number of people take metformin or rapamycin in the hope of buying themselves extra years. Beneath all of this is a question that biology has not yet answered cleanly: What actually controls the rate at which we age?
We know lifespans vary enormously across species. Some animals live for decades while others, like the African turquoise killifish Nothobranchius furzeri, measure their lives in months. But what is tricky to explain is why individuals of the same species, living under the same conditions, age at different rates. Most attempts to answer this have taken a comparative omics approach involving long- and short-lived species. But we do not have a confirmed regulator yet.
A new preprint by Takaochi and colleagues offers one. The authors identified the gut ceramidase Asah2 as a determinant of species- and strain-specific ageing rate in vertebrates, and they did so through a chain of evidence that moves from genomics through genetics through lipid biology to the microbiome.
An unusual fish with an unusual property
Nothobranchius furzeri lives for only a few months; the shortest lifespan of any vertebrate kept in captivity. In that compressed time, it shows neurodegeneration, muscle wasting, and tumours. It is, in effect, a fast-forward model of vertebrate ageing. But what makes this species especially useful for the question at hand are its strains. Different strains show up to twofold differences in lifespan, yet they do not differ in growth rate or age at sexual maturity. They differ specifically in how fast they age. This then serves as an ideal model to identify responsible regulators.
The genetic regions responsible for these lifespan differences had been mapped, but the causative genes were unknown. Takaochi and colleagues, therefore, tried to find them.
Following the clues
Comparing whole-genome sequences of a short-lived strain and a long-lived strain, the authors screened for single-nucleotide variants predicted to damage or inactivate proteins. They narrowed the list to tissue-specific metabolic enzymes based on the reasoning that these would be traceable targets for intervention. They finally arrived at six candidates. From these, one stood out: Asah2, encoding a neutral ceramidase expressed specifically in intestinal epithelial cells. In the long-lived strain, a single-nucleotide deletion eliminates Asah2 protein expression entirely.
The cross-species picture added weight. Analysing 464 vertebrate species, the authors found that several relatively long-lived species within their taxonomic class lack amino acid residues essential for Asah2 enzymatic activity.
What knocking out Asah2 does
The result was clear. In male fish, asah2 KO extended lifespan and attenuated age-related decline in locomotor activity. Tissue analysis showed the effects were systemic with
- Reduced protein aggregation in the cerebellum
- Less muscle fibre atrophy
- Fewer senescent cells in the liver and intestine
- Stronger intestinal barrier integrity
- Suppressed age-associated inflammation.
One enzyme, in one tissue, appears to slow ageing across the body (Figure 2).
The effect was male-specific. Female lifespan was not extended, a finding the authors link to the observation that Asah2 expression rises with age in male but not female N. furzeri. Whether this reflects genuinely dimorphic ageing biology or protection via alternative routes is not yet clear, but it is a finding that deserves attention.
Ceramides; not just for skincare
Here is where the story gets interesting for anyone who has glanced at a moisturiser label. Ceramides are a class of lipid well known in dermatology for their role in skin barrier function. They are a common ingredient in creams marketed for hydration and anti-ageing. What this paper shows is that specific ceramide species in the intestine may do something more fundamental.
Because Asah2 degrades ceramides, its loss predictably raises ceramide levels. The authors found that asah2 KO elevated two specific ceramide species, Cer(d16:1/22:0) and Cer(d17:1/22:0), in the intestine, without changing total ceramide content. They then fed these ceramides directly to wild-type fish. Ceramide supplementation alone extended lifespan, preserved muscle, and reduced intestinal inflammation. The ceramides are not just a readout of Asah2 loss; they are doing the work.
The microbiome connection
Over the years, evidence has mounted for the influence of the gut on our health. (Daniel et al., 2021). In this context, a closer look at Asah2 revealed that it sits on the luminal side of intestinal epithelial cells, facing the gut. Wild-type fish showed significant shifts in microbiota composition between 2 and 4 months of age; asah2 KO fish did not. The knockout showed enrichment for Lactobacillus, a genus that declines with age in humans, and for bacteria associated with short-chain fatty acid production, metabolites known to support intestinal barrier integrity and reduce systemic inflammation.
To test causality, the authors transplanted microbiota from young asah2 KO donors into young wild-type recipients. The recipients showed reduced brain protein aggregation and intestinal inflammation at 4.5 months, recapitulating aspects of the KO phenotype through the microbiome alone.
The proposed mechanism runs: Asah2 activity -> intestinal ceramide levels -> microbiota composition ->systemic ageing rate.
Why is this exciting?
- Humans have the Asah2. Intact. In the intestine.
- Intestinal Asah2 expression rises with age, and a UK Biobank analysis found that higher offspring plasma Asah2 levels were associated with shorter parental survival.
The authors acknowledge the limitations: offspring ASAH2 was measured, not parental intestinal levels. But the directionality is consistent with the fish data.
Why this matters
What I find compelling about this paper is that it offers a molecular handle on something the field has long suspected but struggled to pin down: a gut-to-soma ageing axis with a specific, tractable mediator. The ceramide supplementation result is particularly striking. It suggests that dietary intervention, not genetic manipulation, might be sufficient to recapitulate the benefit. Whether that holds in longer-lived vertebrates remains to be seen, but it is the kind of result that opens doors.
The sex-specificity is a thread worth pulling. A protective mechanism that operates in males but not females is not a limitation. It is a biological signal, and one that has implications for how we think about sex differences in ageing more broadly. They do say ‘Women age faster’.

Questions for the authors
- The microbiota transplant used young donors and young recipients, with readouts at 4.5 months. Could transplantation into already-aged fish still attenuate ageing phenotypes, or is the microbiota effect primarily preventive?
- The ceramide supplementation was given for four weeks at 1.5-2.5 months of age. Is there a critical window, or would later intervention still be effective?
- Given the sex-specific effects in N. furzeri, do male and female fish differ in baseline intestinal ceramide levels or microbiota composition in ways that might explain the female insensitivity to asah2 KO?

Peer Review in Biomedical Sciences
Communication of scientific knowledge has changed dramatically in recent decades and the public perception of scientific discoveries depends on the peer review process of articles published in scientific journals. Preprints are key vehicles for the dissemination of scientific discoveries, but they are still not properly recognized by the scientific community since peer review is very limited. On the other hand, peer review is very heterogeneous and a fundamental aspect to improve it is to train young scientists on how to think critically and how to evaluate scientific knowledge in a professional way. Thus, this course aims to: i) train students on how to perform peer review of scientific manuscripts in a professional manner; ii) develop students' critical thinking; iii) contribute to the appreciation of preprints as important vehicles for the dissemination of scientific knowledge without restrictions; iv) contribute to the development of students' curricula, as their opinions will be published and indexed on the preLights platform. The evaluations will be based on qualitative analyses of the oral presentations of preprints in the field of biomedical sciences deposited in the bioRxiv server, of the critical reports written by the students, as well as of the participation of the students during the preprints discussions.

Keystone Symposium on Stem Cell Models in Embryology 2026
The Keystone Symposium on Stem Cell Models in Embryology, 2026, was organised by Jun Wu (UT Southwestern), Jianping Fu (University of Michigan) and Miki Ebisuya (TU Dresden) and held at Asilomar Conference Grounds in California (US).
